Hypoperfusion (reduction of blood flow) can lead to hypoxia (concentration of oxygen falls below life sustaining levels) in the brain, which can lead to cell death and abnormal brain function. In theory, stem cell therapy may reverse hypoxia while activating the brain's "self repair mechanisms."

The connection between immune function and nervous system is strong. Stem cells may repair the immune system, which in theory would lead to improved neurological functioning.

Obama overturns Bush policy on stem cells – CNN.com

WASHINGTON (CNN) -- President Obama signed an executive order Monday repealing a Bush-era policy that limited federal tax dollars for embryonic stem cell research.

President Obama signs the executive order on stem cell policy Monday at the White House.

Obama's move overturns an order signed by President Bush in 2001 that barred the National Institutes of Health from funding research on embryonic stem cells beyond using 60 cell lines that existed at that time.

Obama also signed a presidential memorandum establishing greater independence for federal science policies and programs.

"In recent years, when it comes to stem cell research, rather than furthering discovery, our government has forced what I believe is a false choice between sound science and moral values," Obama said at the White House.

"In this case, I believe the two are not inconsistent. As a person of faith, I believe we are called to care for each other and work to ease human suffering. I believe we have been given the capacity and will to pursue this research -- and the humanity and conscience to do so responsibly." Watch as Obama announces he's lifting the funding ban

The president pledged to develop "strict guidelines" to ensure that such research "never opens the door to the use of cloning for human reproduction."

Such a possibility, he maintained, is "dangerous, profoundly wrong and has no place in our society or any society."

Obama's order directs the NIH to develop revised guidelines on federal funding for embryonic stem cell research within 120 days, according to Dr. Harold Varmus, president of Memorial Sloan-Kettering Cancer Center and co-chairman of Obama's science advisory council.

"The president is, in effect, allowing federal funding of human embryonic stem cell research to the extent that it's permitted by law -- that is, work with stem cells themselves, not the derivation of stem cells," Varmus said in a conference call with reporters Sunday.

Researchers are closer to unraveling the mystery of how Timothy Ray Brown, the only human cured of HIV, defeated the virus, according to a new study. Although the work doesnt provide a definitive answer, it rules out one possible explanation.

Brown remains one of the most studied cases in the HIV epidemics history. In 2006, after living with the virus for 11 years and controlling his infection with antiretroviral drugs, he learned that he had developed acute myeloid leukemia. (The leukemia has no known relationship to HIV infection or treatment.) Chemotherapy failed, and the next year Brown received the first of two bone marrow transplants a common treatment for this cancer and ditched his antiretrovirals. (An American then living in Berlin, Brown has been known to researchers for years as the Berlin patient.

When HIV-infected people stop taking these drugs, levels of HIV typically skyrocket within weeks. Yet researchers scouring Browns blood over the past seven years have found only traces of the viral genetic material, none of which can replicate.

Today, researchers point to three factors that might independently or in combination have ridden Browns body of HIV. The first is the process of conditioning, in which doctors destroyed Browns immune system with chemotherapy and whole-body irradiation to prepare him for his bone marrow transplant.

Second, his oncologist, Gero Htter, took an extra step that he thought might not only cure the leukemia but also help rid Browns body of HIV. He found a bone marrow donor who had a rare mutation in a gene that cripples a key receptor on white blood cells that the virus uses to establish an infection.

The third possible explanation is that Browns new immune system attacked remnants of his old one that held HIV-infected cells, a process known as graft vs. host disease.

In the new study, a team led by immunologist Guido Silvestri of Emory University in Atlanta designed an unusual monkey experiment to test these possibilities.

Bone marrow transplants work because of stem cells. Modern techniques avoid actually aspirating bone marrow and instead can sift through blood and pluck out the stem cells needed for a transplant to engraft. So the researchers first drew blood from three rhesus macaque monkeys, removed stem cells and put the cells in storage. They then infected these animals and three control monkeys with a hybrid virus, known as SHIV, that contains parts of the simian and human AIDS viruses. All six animals soon began receiving antiretroviral drugs, and SHIV levels in the blood quickly dropped below the level of detection on standard tests, as expected.

A few months later, the three monkeys with stored stem cells underwent whole-body irradiation to condition their bodies and then had their own stem cells reinfused. After the cells engrafted, a process that took a few more months, the researchers stopped antiretrovirals in the three animals and in the three controls. SHIV quickly came screaming back in the three controls and two of the transplanted animals. (One of the transplanted monkeys did not have the virus rebound, but its kidneys failed and the researchers euthanized it.)

Stem cells are used extensively in Europe and Asia to treat these conditions.

We have anecdotal and experimental evidence that stem cell therapy is effective in healing and regeneration. Stem cells seek out damaged tissues in order to repair the body naturally. Co-workers honor doctor by registering as bone marrow donors

PASCAGOULA, MS (WLOX) -

Nearly 120 doctors, nurses, technicians and other Singing River Health System employees lined up to register as a bone marrow donor in honor of their co-worker, Dr. Jeremy Simpler, who is battling cancer.

"I don't want to get emotional, but very, very devastating when we found this out because he is wonderful. He's a people person. He treated us like family," Singing River Health System Surgical Tech Patricia Taylor said.

A stranger, who gave 40 seconds to get his or her cheeks swabbed, could end up saving Simpler's life. Now, his co-workers want to repay that favor.

"Every day, we have someone looking for a match," Mattie Coburn, with the Mississippi Marrow Donor Program said.

Coburn said 70 percent of patients who need a transplant rely on the registry, because they do not have a match in their family.

There are two different ways donors are asked to donate.

"Bone marrow transplant, it is outpatient. You are put to sleep under anesthesia. We are going to go to the hip and pelvic bone with a sterile needle and syringe," Coburn said. "You are not going to feel it. We keep you overnight for observation, and release you with a Band-Aid over where the needles were. PBSC, peripheral blood stem cell, is similar to giving platelets. We draw blood, separate the cells, you get your blood back."

Three years ago, Singing River Health System Dr. Clinton Hull donated bone marrow.

"It was a really good feeling," Hull said. "The last communication I had through the bone marrow service was the patient had returned to their daily activities and living, so that makes me feel really good."

A review of more than 400 treatment cases shows 60% to 70% of patients with multiple sclerosis (MS) undergoing stem cell transplants had a 3-year progression-free survival, with treatment-related morbidity of 1% to 2%. In vitro studies also indicate an end to MS causative autoimmune reactions, with clinical remission persisting after full normal immune reconstitution. Prospective randomized trials for stem cell treatment of MS were started in 2008 in Europe and the USA.

MS, also known as disseminated sclerosis or encephalomyelitis disseminata, is a chronic inflammatory and degenerative disease of the central nervous system (CNS). Autoimmunity against myelin sheaths is observed in MS and leads to demyelination of neurons. Loss of myelin and plaque formations (scleroses) in white matter of the CNS disrupts neural signaling, producing physical and cognitive impairment. The cause remains unknown.

Stem cells used for the treatment of MS are derived from adult bone marrow. These hematopoietic stem cells (HSCs) are multipotent in nature. HSCs are capable of self-renew and differentiation into cells of all blood lineages. Transplanted HSCs can also differentiate into specific cell types (e.g., muscle, skin, liver, lung, and neural cells).

Pharmaceutical treatments for MS focus on the inflammatory mechanisms of the disease. These treatments are of limited efficacy in suppressing immune-mediated CNS inflammation and clinical relapses. After promising studies in animal models of autoimmune diseases, immunosuppression followed by HSCs was tested as treatment in severe forms of MS.

The benefits of HSC therapy for MS include HSCs entering the brain and producing new neural cells. Remyelination has also been observed. HSC transplantation is also associated with changes in thymic output. These changes include a new and diverse T-cell receptor (TCR) repertoire.

HSC transplantation can promote CNS repair and regeneration via different modes of action. Proposed mechanisms include cell replacement, remyelination, and bystander activity (immunomodulation). Inflammatory activity is also reduced by treatment. Two prospective randomized trials are in progress in Europe and the USA. This studies are the first to clinically test if stem cell transplantation is superior to immunosuppressive treatments in severe cases of MS.
